We report two phosphorescent sensors for biological copper (ZIr2) and zinc ions (ZIrF). The sensor constructs are heteroleptic Ir(III) complexes having two chromophoric ligands, thereby affording dual phosphorescence emission in metal-free states. Metal-chelating di(2-picolyl)amino group is tethered to the Ir(III) complexes. The probes exhibit ratiometric responses to copper(II) and zinc ions, while photophysical processes to the sensing behaviors are different. Extensive studies including steady-state and transient photophysical measurements, electrochemical experiments, and quantum chemical calculations were performed to establish sensing mechanisms. Analytical parameters such as Kd and pKa have been determined in buffered solutions at pH 7.0. Due to the long-lived phosphorescence, the sensors offer additional advantages, such as time-gated acquisition and photoluminescence lifetime sensing. In vivo bioimaging abilities of the sensors have been verified.
