Nucleic acid aptamers are potentially useful for targeted delivery of drug-encapsulated liposome particles into cell or tissue of diseases. In this study, we report the design of aptamer-conjugated liposomes (named as aptamosomes) that achieve selective and efficient delivery of anticancer reagent using the cancer cell specific RNA aptamer. Specifically, we prepared the prostate cancer cell-specific aptamosomes conjugated with the RNA aptamer specific to PSMA (prostate-specific membrane antigen) highly expressed on cancer cell surface. The PSMA RNA aptamer was extended for annealing with complementary DNA linker which was labeled with thiol group and fluorophore (FITC). The modified thiol group of DNA linkers were conjugated with maleimide group on the surface of PEGylated micelles. The aptamosomes of nano scale (90~100nm) prepared by the post-insertion method specifically bind to the PSMA-positive LNCaP prostate epithelial cells not to the PSMA-negative cancer cells, as compared with nontargeted nanoparticles that lack the PSMA aptamer. Thus, we have demonstrated that doxorubicin-encapsulated aptamosomes were significantly more cytotoxic to the targeted LNCaP cells than to the nontargeted PC3 and A549 cells.
