Contamination of the environment with heavy metal ions in aqueous solution has been an important worldwide concern for decades. Especially mercury ions act as environmental pollutant and they have serious medical effects. Therefore, the rapid and sensitive analysis of mercury ions in water or food is highly important. We report a simple and sensitive detection of mercury ions using thrombin and thrombin aptamer. Thrombin is a protein that plays many roles in the coagulation cascade-converting soluble fibrinogen into insoluble strands of fibrin as well as catalyzing many other coagulation-related reactions. Aptamer is short single-stranded oligonucleotide that fold into three-dimensional structures that allow them to bind a specific protein. Exposure of aptamer-modified surface to mercury ion converts flexible aptamer into hairpin structure due to the formation of T-Hg2+-T, which makes it difficult for thrombin to interact with aptamer. Through this presentation, we demonstrate that the our system enables ultrasensitive naked eye detection of mercury ion.
