Thrombin is a protein that plays many roles in the coagulation cascade-converting soluble fibrinogen into insoluble strands of fibrin as well as catalyzing many other coagulation-related reactions. Therefore, a sensitive detection of thrombin in blood is of great interest and importance. To detect thrombin, we use the thrombin binding aptamers. Aptamers provide several advantages such as good stability, simple synthesis, high resistance against denaturation, and high sensitivity. When the solid surfaces modified with the aptamer is exposed to a thrombin solution, thrombin is immobilized onto the surfaces. The thrombin-immobilized surfaces are able to convert a soluble fibrinogen into an insoluble strand of visible fibrin, where the amount of fibrin on the surfaces depends on the concentration of thrombin in the solution. In this presentation, we demonstrate that our method permit a highly sensitive thrombin detection through only naked eye without instruments.
