We address here the nature of A5 binding on PS-containing lipid membrane at the single molecular level. A5 is a PS lipid-binding protein forming a tightly bound A5-Ca2+-PS complex, and its binding affinity between A5 and PS was previously known to vary greatly depending upon phosphatidylserine (PS) and calcium ion concentrations at the many molecular level. In this work, with single molecule force spectroscopic technique, we measured precisely the binding strength of single A5-(Ca2+)x(PS)y complex in controlled environment. The observed unbinding force was approximately constant even when the PS concentration was varied in the range of 25% ~ 80% mole fractions. It was also observed that after A5 binding, the lipid membrane became stiff and even condensed to some degree. Our data shows that the number of PS molecules to take part in the complex formation is more or less constant as long as the PS concentration is higher than 25%, and the high-density lipid domain can form after the A5 binding to the PS membrane.
