Every cell has different level of proteins even if their genomes are exactly the same. This causes a phenomenon called “gene expression noise” and it implies significant cell-to-cell variations. Depending on the origins, noise is classified into intrinsic and extrinsic noise. Normally extrinsic noise has various sources, and therefore it is difficult to model it. The number of RNA polymerase (RNAP) has been expected for a long time as one of the major noise sources. To measure the level of RNAP concentration and noise, we established a bacterium system, in which fluorescently labeled RNAPs are expressed and these RNAPs generate other two types of fluorescent proteins. From this work we found that RNAP concentration has major effect only on the extrinsic noise, not on the intrinsic noise. The extrinsic noise was dropped when RNAP concentration was tightly controlled, which means that RNAP concentration has major effect on the extrinsic noise. Our result shows that RNAP has a significant role on determining the cell-to-cell variation.
