2-Pyrones are important structural subunits in a wide variety of biologically active natural products, as well as useful versatile synthetic intermediates. Recently, the activity of 2-pyrones as potent HIV protease inhibitors invoked additional interest in the investigation of 2-pyrones and their analogues. Phosphorus 2-pyrones are particularly interesting due to their similarity in reactivity and bioactivities with the carbon species. However, until 2004, only five phosphorus 2-pyrone analogues have been reported in the literature. Unfortunately, their methods have not been thoroughly examined and they are usually lengthy procedures which suffer from low yields. In 2005, Ding et al. prepared 2H-1,2-oxaphosphorin 2-oxides to show efficient cyclization reaction using Ag2CO3 from (Z)-2-alken-4-phenylethynyl-phsphonic monoesters. However, only six examples were shown and the synthesis of (Z)-2-alken-4-phenylethynyl-phsphonic monoesters is complicated. Therefore, this has led us to develop a novel method for the synthesis of versatile phosphorus 2-pyrone.
