The 5-HT2C receptors appear to be restricted to the central nervous system (CNS) where it demonstrates a wide distribution. The receptors have been considered as therapeutic targets for the treatment of various central nervous system disorders such as depression, anxiety, epilepsy, schizophrenia, and sleep disorders. Also, 5-HT2C receptors have been considered as target receptor for anti-obesity. However, the limit to the development of up to the present does not possess good selectivity and specificity of drug candidates to the receptors. They also activate 5-HT2A and 5-HT2B receptors, resulting in various side effects associating to these receptors, hallucination and valvular hypertrophy respectively. Therefore, anti-obesity drug candidates are greatly focused on developing more selective and specific 5-HT2C agonists. As part of discovering novel 5-HT2C agonists, we designed and synthesized 40 compounds. The synthesized compounds were biologically evaluated against 5-HT2C. The detailed design, synthesis and biological evaluation of 5-HT2C agonists as anti-obesity drugs will be presented.
