Self-assembled monolayers (SAMs) have been widely used as a modification method to immobilize biomolecules. However, the additional layers of SAM and biomolecules make the electron transfer difficult as they cause a large overpotential in electrical signals. Electron transfer mediation is the most popular solution. Ferrocene-undecanethiol was used as an electron transfer mediator due to its electroactive ferrocene moiety. Mercapto-dodecanoic acid (MDA) was used for immobilization of bio-molecules. However, the electron transferability of mixed SAMs is affected by pH, the kind of electrolytes, and the composition and kind of the thiol molecule. Therefore the optimization of the carboxyl acid and ferrocene molecule ratio is the crucial factor for the electrochemical detection. In this study, we found out appropriate ratio of MDA and ferrocene molecule and other conditions for biosensor.
