Surface plasmon resonance ellipsometry(SPR-E) are used to measure interaction between proteins. Measurements are made under conditions of continuous flow, where the biospecific sensor chip surface forms one wall of the flow cell. Microfluidic chip using continuous flow has advantage to the real-time, label-free aspects of the measurement. And, Microfluidic chip system continuously monitors each binding step in the assay procedure, in contrast to many other techniques. Polydimethylsiloxane(PDMS) has advantage to optical transparency in a broad range of spectrum, low cost, and simple processing using the micro molding technique. But, molding technique is difficult to materialize 3-d structure of chip. We developed the 3-d PDMS microfluidic chip for SRP-E system using molding technique. We used laser micromachining technique for 3-d structure of chip. That is, we fabricated two types of microchannel structures in PDMS, and two PDMS plates which are bonded together with a single PDMS alignment.
