Synthesis of porphyrin-incorporated peptide nanofibers: A possible optical probe for DNA hybridization

A biosensor based on porphyrin –incorporated peptide nanofibers has been prepared to detect DNA. We synthesized this material by coupling of D,D- diphenyl alanine peptide with trans-Dihydroxo [5,10,15,20-tetrakis(p-tolyl) porphyrynato] tin(IV)(SnTTP) porphyrin under hydrothermal reaction condition at 115oC. Using atomic force microscopic technique we found that the obtained material have fiber -like morphology. From absorption spectra we observed the porphyrin –modified peptide nanofibers were red shifted compared to free monomer porphyrin .This may be due to the J-aggregation of porphyrin inside the peptide nanotube during intercalation. Upon interaction of these nanofibers with DNA, the complementary DNA sequence showed higher fluorescence than non complementary DNA sequences. It was also found that this material showing higher life time upon interaction with complementary DNA. These results indicate that this matrial can be applied in future for the analysis of DNA and specific targeting of protien.
