Chip-Based Energy Transfer System between Quantum Dots and Gold Nanoparticles for Multiplexed Assay of Proteases

We developed a multiplexed assay system of proteases (matrix metalloproteinase, caspase, and thrombin) by measuring the energy transfer between quantum dot (QDs) and gold nanoparticles (AuNPs) on glass. To construct a chip-based system, biotinylated peptide substrate was conjugated to monomaleimide-functionalized AuNPs, and the resulting AuNPs were associated with streptavidin-conjugated QDs (SA-QDs) that had been deposited on a glass slide. In this system, while the photoluminescence of different donor QDs were quenched due to the presence of a common energy acceptor (AuNP) in close proximity, the protease activity caused specific modulation in the efficiency of the energy transfer between the acceptor and donor, thus enabling the multiplexed protease assay. Our chip-based format will find applications to diagnosis of protease-related diseases and screening of potential drug inhibitors with high sensitivity in a high throughput way.
