Preparation of anionic liposomes coated with albumin and their plasma protein binding chracteristics

Liposomes, nano-sized vesicles composed of lipid bilayers, have been used as drug carriers. Howerer, rapid clearance of plain liposomes by the reticuloendothelial system (RES) limits their circulation in bloodstream and targetability to pathological tissues or cells. In this study, liposomes of which surfaces was coated with bovine serum albumin (BSA) were prepared by conjugating BSA molecules to anionic liposomes via electrostatic interaction at lower pH than isoelectric point of BSA. Mean particle diameter and zeta potential value of BSA-coated liposomes (BCL) were 119.0±7.2 nm and -7.3±3.9 mV, respectively. BCL inqubated in blood plasma showed lower increment of mean particle diameter due to less adsorption of plasma protein onto their surface compared with plain or anionic liposomes. These results suggest that BCL could increase their circulation time in bloodstream because of inhibiting adsorption of opsonin proteins to the liposomes.
