The human replication protein A (RPA) is a heterotrimeric protein complex. It consists of 70, 32, and 14 kDa subunits that are involved in DNA replication, repair, and genetic recombination. Our previous study showed that RPA contains a 4-cysteine type Zinc-finger motif which is not essential for RPA’s DNA binding activity. However it is involved in the regulation of DNA binding activity of RPA through reduction-oxidation (redox). To find out the roles of this zinc finger motif in RPA, we performed a series of experiments including the Circular Dichroism (CD) Spectroscopy and the electrophoretic mobility shift assay (EMSA). The results showed that redox-change could not cause the change of the secondary structure of RPA. In contrast, the loss of Zn2+ caused the change of tertiary structure of RPA. These results revealed that Zinc finger motif play a very important role in structural change of RPA.
