MBHA (4-methylbenzhydrylamine) resin has been widely used for the synthesis of carboxamide or peptide amides and its industrial demand as a solid support for the production of peptide drugs is increasing. In contrast to the previous methods, we prepared MBHA resin in short reaction steps by benzotriazole-catalyzd amidoalkylation. First, formamido PS resin, the precursor to MBHA resin, was synthesized from 1% DVB-PS resin using 4-tolylidene-bis(formamide) and benzotriazole. By removing formamido group by partial hydrolysis, a core-shell type MBHA resin was obtained efficiently. The core-shell type structure of the MBHA resin was analyzed by two-photon microscopy and it synthetic performance in Solid-Phase Peptide Synthesis (SPPS) was also evaluated.
