Nanobiotechnology that applies nano-sized materials to biological system has been rapidly developed. Since the magnetic nanoparticles can be controlled in size they have become an important material in biology and biomedicine. ln the recent experiments, we achieved to isolate specifically cysteine-containing peptide from the complex peptide mixture including human serum by using silica-coated magnetic nanoparticles that were surface-modified with SPDP. Using these SPDP modified nanoparticles that react toward free thiol of cysteine, magnetic nanoparticles were biofunctionalized with biomolecules containing cysteine. These biofunctionalized nanoparticles were able to purify target proteins fused with specific protein that recognize biomolecules as own substrate and bind via enzyme-substrate interaction. The purified proteins were digested and analyzed by LC/MS/MS experiments.
