Phospholipids are important constituents of all living cell membranes. In this study, we have analysed phospholipids. The phospholipid structures have variations in substitution at each of the three carbon atoms. Using normal-phase HPLC to first separate the phospholipids by class will eliminate the issue of the molecular species of TAGs, PEs, PCs co-eluting in positive ion mode and PIs, PEs, PCs in negative ion mode co-eluting. When the reversed-phase column was used, the PLs could be separated depending on their hydrophobicity, essentially the length of their fatty acyl chains and the number of unsaturated bonds in them. In this study, we have tried to assign 2-Dimensional LC-MS, off-line normal-phase and reversed-phase LC separation in tandem mass spectrometry method for phospholipids analysis. We have assigned 2-Dimensional LC-MS lipids profiling method based on off-line tandem MS.
