NS1 protein encoded by influenza A viruses (NS1A protein) binds the viral dsRNA that are produced during infection, thereby blocking the activation of at least one cellular protein (PKR kinase) that would otherwise shut down the synthesis of viral proteins. Discovery of novel agents that inhibit the binding of NS1A with dsRNA and have druglike properties is therefore a major focus in academics and industrial laboratories. In this study, we describe the development and optimization in a 384-well format assay for the identification of NS1A inhibitors using fluorescence polarization. A primary screening of 4,000 compounds was performed and 15 hits have been identified. The authors also present detailed characteristics of the assay, including its dynamic range and Z'-factor values, which indicate that this robust assay is well suited for kinetic and inhibition studies in HTS formats.
