Synthetic Studies of Tail-Modified KRN7000 Analogues

Since its first discovery by Kirin Pharmaceuticals, an α-galacotosyl ceramide named KRN7000 has received much attention. The subsequent investigations of the biological activity of KRN 7000 have revealed remarkable physiological activities against several diseases such as cancer, malaria, juvenile diabetes, hepatitis-B and antoimmune diseases such as encephalomyelitis. The bioactivity and bioavailability of KRN7000 are expected to depend on its tails. Although sphinganine core-containing KRN7000 has shown similar potency and types of activation to KRN7000, research on the variation of its tail has not yet been done. Research on the stereochemical variation of the phytosphingosine core has not been thoroughly carried out either. For these goals, we have synthesized KRN7000 analogues, using D-erythro sphinganine, L-threo sphinganine and D-lyxo phytosphingosine as the core, and aromatic ring containing chains as the tail. The stimulatory activity of these KRN7000 analogues is currently being evaluated in iNKT cells.
