Galanthamine has potent inhibitory activity against acetylcholinesterase(Ache) in CNS, and has been approved for the treatment of Alzheimer’s patients through the enhancement of their cognitive functions. As a part of our ongoing research program on 3,5-dibromo-2-pyrone as novel enophile synthons, we have explored its potential applicability to the target-oriented synthesis with the maximum use of the resultant densely functionalized cycloadducts. We have recently achieved the total synthesis of galanthamine, using the regioselective coupling reactions of 3,5-dibromo-2-pyrone and IMDA as key steps. Presented herein is our 2nd-generation synthesis of galanthamine, more suited for large-scale preparation.
