pKa, logP, lipophilicity, solubility and permeability are the key properties to evaluation of Hit compounds from the high-throughput screening(HTS). Pre-formulation assay system can greatly attribute to national drug discovery programs. Solubility is important for proper interpretation of in vitro profiling results. Early screening of physicochemical and ADME properties has become crucial in drug discovery to address high attrition during drug development. The information of these properties is used to design, select and optimize leads for the highest chances of success and insures that discovery experiments are conducted under optimal conditions. To address these problems, Korea Chemical Bank(KCB) established physicochemical property evaluation and pre-formulation assay system. PAMPA system was settled for evaluating permeability. pKa and logP values can be estimated using titration method of GLpKa/D-PAS, and solubility is measured from the nephelometry experiments. To verify the integrity of the compounds, LC/MS and UPLC analysis is performed.
