An all solid state potentiometric immunosensor has been developed to study the activation process of neuronal nitric oxide synthase (nNOS). Simple drop-coating of carboxylated poly-vinyl chloride (PVC-COOH) membrane containing H+ ionophore was performed on a glassy carbon electrode. Antibody of phospho-nNOS IgG was covalently bonded on the PVC. After the formation of the antibody-antigen immunocomplex upon adding nNOS antigen, the immunosensor was further incubated in secondary antibody anti-IgG urease conjugate solution. The amplified detection of the immunocomplex by urease mediated conversion of urea into ammonia associated with pH increase uses potentiometry as a transduction method. Experimental parameters such as the amount of nNOS antigen immobilized on the electrode surface and pH variation responses will be presented in detail.
