S-Nitrosothiols (RSNOs), a group of storage and delivery vehicles of nitric oxide (NO) in the body regulate several biological processes, including vasodilation, platelet activation, neurotransmission, and tissue inflammation. Small-molecule RSNOs such as S-nitroso-N-acetyl-DL-penicillamine (SNAP), S-nitroso-N-acetyl-L-cysteine (SNAC), and S-nitrosoglutathione (GSNO) have been extensively investigated as conveyance for NO with well-documented antimicrobial and antithrombotic effects. Various analytical methods have been developed for the detection of RSNOs. Recently, amperometric approaches to determine RSNOs has been reported by measuring NO generated from RSNOs. Indeed, RSNOs are readily decomposed by an organometallic catalyst (e.g., copper(II) and organoselenium derivatives), and thus produce electrochemical measurable NO. However, previous RSNO sensors have a difficulty in distinguishing between pre-existed NO and NO generated from RSNO. To overcome this problem, we herein report on an optically switchable method for an amperometric NO sensing by using a optical fiber. When the light is out, the sensor measures only pre-existed NO. On the other hand, when the light is on, the sensor measures both NO, i.e., pre-existed NO and NO generated from RSNO. Of note, RSNOs are also photolytically decomposed. Thereby the sensor makes a distinction between NO signal and decomposed NO. The new RSNO sensor is applied to detect various RSNOs (e.g., S-nitroso-N-acetyl-DL-penicillamine, S-nitroso-N-acetyl-L-cysteine, S-nitrosoglutathione and S-nitroso-L-cysteine). In order to optimize the new method, experimental factors such as light intensity and exposing time are changed and the effect of changes in the sensor characteristics will be studied. Furthermore, the developed RSNO sensor are employed to determine RSNOs in human blood serum.
